Cytotoxic T cell killing assay

Explicyte offers to evaluate the ability of candidate compounds to modulate the effector function of immune cell subpopulations that mediate tumor cell apoptosis. This cytotoxic T cell killing assay monitors both cell death and cell proliferation of cancer cells in real time, in the presence of activated immune cells.

Assay Principle

 

  • Target tumor cells are co-cultured with immune cells (e.g., purified T cells, PBMCs) activated to promote tumor cell killing.
  • Tumor cell count is enabled by labeling target cells with a range of NucLightTM Live-Cell Labeling Reagents. Tumor cell death is measured in real-time by adding the mix-and-read IncuCyteTM Caspase 3/7 Reagent to the cultures.
  • Automated image analysis enables selective quantitation of tumor cell count and cell death in the continuous presence of lymphocyte populations and without separation or liquid sampling.

Representative Results

T cell killing: immune cell killing of SKOV-3 ovarian cancer cells by activated PBMC.  Real time detection of apoptotic cells (yellow objects) using Caspase3/7 IncucCyteTM probe. Time courses of cell death in SKOV-3 tumour cells co-cultured by antiCD3 il2 – treated PBMCs.
